ABSTRACT

Objective: To explore the current situation of anxiety, subjective well-being in occupational population and the mediating effect of resilience. Methods: From March 24th to 26th, 2020, a cross-sectional survey was conducted among occupational population aged ≥18 years old using online questionnaires. A total of 2134 valid questionnaires were obtained, with respondents from 30 provinces, autonomous regions, and municipalities directly under the Central Government. Their general demographic data, subjective well-being, anxiety, and resilience were collected. Pearson χ(2) test and Spearson correlation analysis were used for data analysis, and structural equation model was used to explore the mediating effect of resilience on anxiety and subjective well-being. Results: The age of the respondents ranged from 18 to 60 years old, with an average age of (31.19±7.09) years old, including 1075 (50.4%) women and 1059 (49.6%) men. The positive rates of low subjective well-being and anxiety were 46.5% (992/2134) and 28.4% (607/2134), respectively. Anxiety scores were significantly negatively correlated with subjective well-being scores and resilience scores (r(s)=-0.52, -0.41, P<0.05), while resilience was significantly positively correlated with subjective well-being (r(s)=0.32, P<0.05). Structural equation models showed that anxiety had a negative predictive effect on subjective well-being, while resilience not only had a positive predictive effect on subjective well-being, but also played a mediating role between anxiety and subjective well-being, with a mediating effect of 9.9%. Conclusion: The situation of anxiety and well-being in the occupational population is still not optimistic, and resilience has a mediating effect between anxiety and subjective well-being.

ABSTRACT

Objective During the process of growing cells on coverslips by traditional method , damages like detachment and fragmentation always happen to slips , which affect the experimental process .The article aimed to introduce a new method of growing cells on coverslips, which facilitated experimental operation and simplified the experiment procedure .Methods 1 mL pipette tips were made into cylindrical chambers for use by cutting 20mm from the end.Neutral balsam group: The prepared pipette tips were atta-ched onto the slide by neutral balsam in a symmetrical double row .Glue stick heating group: The glue stick was applied slowly and e -venly on the outer sides along the end of the pipette tips after it was heated .The ends were glued to the slide with the end downward , forming a symmetrical double-row arrangement.Autoclaving was followed after drying and fixing .A comparison was made between the effects of traditional coverslips and hot -melt coverslips.HE staining was used to observe the cell growth . Results The tubes in glue stick heating group were easy to remove from the slides , while the neutral balsam group showed stronger adhesion .During the removing process: there were neutral balsam residues on the slide after the pipette tips were removed in neutral balsam group , while the adhesives could be bluntly removed from the slide in the form of a rubber ring in glue stick heating group .Similar to the traditional meth-od of growing cells on coverdslips , there were no cell contamination or proliferation restriction in the coverslip device made by hot glue .The cells were in good growth and even distribution , without any detachment from the slides. Conclusion The self-made coverslip de-vice by hot-melt adhesive method makes experiment more simplified and convenient , which can be applied in integrated multigroup cov -erslips.Recycling and reusing of consumables during the preparation process save experiment expenses .

ABSTRACT

Governance for health was defined as the activities taken by central government, local authorities, and pertinent departments, to guide the social organizations, enterprises and public, in the pursuit of health and wellbeing, in an integral approach.This article states the fundamental theories of governance for health, and its relationship to“Health China”.It emphasizes that under the leadership of the government, governance for health should establish a platform for participation and a mechanism for equal consultation and cooperation by multi-stakeholders, in order to ensure all participation and all co-sharing.In addition, the article also introduces the concept of universal health management, and illustrates the practical issues needs to be considered in implementing universal health management, from three dimensions of breadth, depth and height.“Breadth” means health management should cover all people in the country and the whole life of people.“Depth” means quality suppliers of health management should provide a list of health management services based on evidence with various packages.“Height” means in order to achieve the effectiveness of health management services, the budget should be secured and shared among the government, the enterprises and the individuals, with agreed percentage.

ABSTRACT

<p><b>BACKGROUND</b>Activation of c-Jun NH(2)-terminal kinase (JNK) has been implicated in neuron apoptosis as well as autophagy in response to various stressors after traumatic brain injury (TBI). However, the underlying molecular pathway remains unclear. Our study assessed whether JNK-mediated p53 phosphorylation might be an important mechanism for enhancing neuron autophagy in response to TBI.</p><p><b>METHODS</b>A total of 186 male Sprague-Dawley (SD) rats (300 - 350 g) were used in this study. By randomized block method rats were randomly divided into four groups: sham-operated (n = 46), TBI (n = 60), TBI + dimethyl sulfoxide (DMSO) (n = 40), and TBI + SP600125 (n = 40). JNK was treated with SP600125, a specific JNK inhibitor. JNK, p-P53, Beclin-1, damage-regulated autophagy modulator (DRAM) and p-bcl-2 were evaluated by Western blotting analysis. The cellular localization and expression of Beclin-1 and DRAM was observed by immunofluorescence and immunohistochemistry, and the expression of Beclin-1-Bcl-2/Bcl-xL complexes was evaluated by immunoprecipitation. Multiple-group comparisons were conducted using analysis of variance (ANOVA). P values of less than 0.05 were considered statistically significant.</p><p><b>RESULTS</b>It was observed that the expression of JNK, p-P53, Beclin-1, DRAM and p-bcl-2 was increasing after TBI, and the expression of Beclin-1 and DRAM was mainly located in the cytoplasm of neurons. But these were significantly inhibited in SP600125 group compared with sham group and TBI + SP600125 group (P < 0.05). The expression of Beclin-1-Bcl-2/Bcl-xL complexes was reduced after TBI.</p><p><b>CONCLUSION</b>JNK-mediated p53 phosphorylation might be an important mechanism for enhancing neuron autophagy in response to TBI.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the effect and potential mechanism of expression of c-jun N-terminal kinase (JNK) signal pathway on neuron autophagy after diffuse brain injury (DBI).</p><p><b>METHODS</b>Male Sprague Dawley rats (n = 216) were randomly divided into four groups: DBI group (n = 54), SP600125 intervene group (n = 54), DMSO group (n = 54) and sham operation group (n = 54). DBI rat model was established according to the description of Marmarou DBI. At different time points (1, 6, 12, 24, 48 and 72 h) after operation, the histopathologic changes of neurons in cortex were observed by HE staining method; The expression of p-JNK, p-P53, DRAM and Beclin-1 were detected by Western blot and immunohistochemistry.</p><p><b>RESULTS</b>The results showed that under light microscope degenerated and necrotic neurons were observed to be scattered in cortex at 6 h after operation in DBI group, but these changes were low in SP600125 intervene group. Compared with SP600125 intervene group, the expression of p-JNK in DBI group were enhanced obviously at 6, 12 and 24 h (F = 17.902, P < 0.05); the expression of p-P53 in DBI group were enhanced obviously at 12, 24, 48 and 72 h (F = 7.107, P < 0.05); the expression of DRAM in DBI group were enhanced obviously at 6, 12, 24, 48 and 72 h (F = 15.455, P < 0.05); the expression of Beclin-1 in DBI group were enhanced obviously at 6, 12, 24, 48 and 72 h (F = 11.517, P < 0.05). Compared with DBI group, the expression of p-JNK, p-P53, DRAM and Beclin-1 in DMSO group were similar at 1, 6, 12, 24, 48 and 72 h (F = 1.509, P > 0.05).</p><p><b>CONCLUSIONS</b>The present results indicate that SP600125 can dramatically improve trauma brain injury from autophagy after DBI and the molecular mechanism is related to the modulation of JNK signal pathway following DBI, while it measures the neuron autophagy by means of intervening JNK signal pathway.</p>

ABSTRACT

Objective To explore the factors affecting blood level of elderly hypertensive patients, using a multilevel analysis model. Methods 927 elderly hypertensive patients from 23 communities were studied, through a multi-stage random sampling method. The influencing factors on systolic blood pressure (SBP) and diastolic blood pressure (DBP) were analyzed through a two-level linear multilevel model, respectively. Results The average blood pressure of subjects appeared as: SBP (139.2±11.7) mm Hg ( 1 mm Hg=0.133 kPa), DBP (85.6±8.6) mm Hg. Ratio of physician versus patients was the factor affecting blood level of subjects from the community level. SBP and DBP of the subjects from the higher physician/patient ratio communities were 3.86 mm Hg and 2.51 mm Hg, lower than the subjects in the lower ratio communities, after controlling the other related factors. Age, gender,overweight/obesity were the individual risk factors of hypertension, while factors as regularl medicine taking, reducing salt intake and related self-efficacy to manage disease could reduce the blood pressure.Reducing salt intake could lower the SBP for 2.44 mm Hg and DBP for 2.03 mm Hg, after controlling the other factors. Conclusion Multilevel analysis model could effectively analyze the hierarchically structured data while both factors from the community and individual levels could affect the blood level among elderly patients with hypertension.

ABSTRACT

Objective To explore the application of repeated measurement‘analysis of variance'(ANOVA)in evaluating the effectiveness of 'community-based hypertension self-management program'.Methods A community-based parallel controlled trial was conducted among 3 communities.169 patients in intervention group took part in the course on hypertension self-management program once a week and 204 patients in control group received routine hypertension management services.Data collected through questionnaire at baseline and 6 months,12 months after intervention and were compared through repeated ANOVA measurement.Results Subjects in the intervention group showed statistical significance and linear trends in health self-evaluation,distress,in lOW spirit,self-efficacy in managing symptoms(SEMS),self-efficacy to managing diseases in general(SEMDG),systolic blood pressure(SBP)and diastolic blood pressure(DBP)over time by univariate test of repeated measurement ANOVA.The score of SEMS increased from 6.84±2.53 at baseline to 8.20±1.44 at 12 months after intervention while SEMDG from 7.28±2.45 to 8.89±1.05,and SBP decreased from 137.66±7.30 inln Hg(1 mm Hg=0.133 kPa)to 130.41±7.71 mm Hg.DBP decreased from 84.13±6.70 mm Hg to 81.04±5.98 mm Hg respectively.Only tow spirit and SBP changed over time were seen in the control group.Self-evaluation,distress,in low spirit,caused by diseases,SEMS,SEMDG and SBP were statistically different between control and intervention groups,and the effect of interaction between groups and time span were statmtically significanton indicators as self-evaluation,low spirit,self-efficacy in managing symptoms,seLf-efficacy tO manage diseases and sBP etc,by multivariate test of repeated measurement ANOVA. Conclusion Repeated measurement ANOVA not only could be used to analyze group-effect,but could also explain the effect and the interaction among groups and time,to make the results more reliable.The self-management approach could improve the health status and self-efficacy of the patients,so as to reduce the blood pressure.Our result showed that it was effective for hypertensive patients to be on the chronic diseases selfmanagement program.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the effect of N-acetyl L-cysteine (NAC) on expressions of matrix metalloproteinases-2, 9 (MMP-2, MMP-9) in lung fibroblasts of SiO(2) exposed rats.</p><p><b>METHODS</b>Seventy-five Wistar rats were divided randomly into three groups. The control group was administered with normal Saline. The model group and the interventional group were administered with SiO(2), and the interventional group was administered with NAC before SiO(2) was administered. Lung fibroblasts were isolated on day 1, 3, 7, 14, 28 after exposure to SiO(2). The expressions of MMP-2 and MMP-9 were evaluated by Immunocytochemistry and RT-PCR.</p><p><b>RESULTS</b>(1) The expressions of protein and mRNA of MMP-2 in the model group were higher than that in the control group on all days after exposure to SiO(2) (P < 0.01). The expression of protein of MMP-9 was higher than the control group on day 1, 3, 7, and mRNA was higher on day 1, 3 (P < 0.01). (2) The expression of protein of MMP-2 in the interventional group was lower than the model group on all days, higher than the control group on day 3, 7, 14, 28, and the expression of mRNA was higher than the control group, lower than the model group, on all days (P < 0.05 or P < 0.01). The expression of protein of MMP-9 in the interventional group was lower than the model group on day 1, 3, 7, but higher than the control group on day 3, 7, and mRNA was lower than the model group on days 1, 3, higher than the control group (P < 0.05).</p><p><b>CONCLUSION</b>NAC inhibits the expressions of MMP-2, MMP-9 in lung fibroblasts.</p>
